The operation process of batch fermentation

Batch fermentation refers to the batch operation of the bioreactor. During the fermentation process, except for the continuous input of air (aerobic fermentation) and the addition of acid and alkali to adjust the pH of the fermentation broth, there is no material exchange with the outside world. This method is simple to operate and is the most commonly used fermentation method.


The main feature of batch fermentation is that all process variables change over time. Including: bacteria, substrates, oxygen supply, products and carbon dioxide release, etc.

The main bioreactor and seed fermentation tank are the main equipment of the whole process. They are all deep culture tanks and are equipped with culture medium, steam sterilization, cooling, ventilation, air filtration, stirring and defoaming devices. Obtain the desired product in the main bioreactor.

Shake flask culture and seed fermentation tanks sometimes use secondary or above secondary culture methods. From shaking flask culture to seed culture to fermentation main culture, it is generally expanded step by step with 10 times the volume.

Process flow of batch fermentation operation:

1. Start work from seed cultivation, firstly activate production strains, expand cultivation in the laboratory, and expand cultivation in seed tank fermentation. 

Including the seed tank high-pressure steam sterilization empty tank, then adding the culture medium for sterilization, and then inoculating the fermented strains which cultivated in the shake flask to seed tank.


2. At the same time that the seed tank starts to carry out the fermentation culture, prepare the main bioreactor in the same way. For large-scale fermentation tanks, the medium is generally not sterilized in the tank, but a special sterilization device is used to continuously sterilize the medium. 

During the cultivation process, the fermentation temperature and pH of the fermentation broth need to be controlled. For aerobic microorganism fermentation, aeration and stirring are also required.


3. When the concentration of bacteria in the seed fermentation tank reaches the specified concentration, inoculate the seed liquid to the main bioreactor. According to the prescribed fermentation conditions, control the fermentation factors, and finish the fermentation process when the fermentation reaches the end conditions. 

The culture solution with accumulated fermentation products is transported to the extraction and refining workshop for downstream processing.

Fermentation cycle refers to the whole process of each batch of fermentation reaction, that is, the sum of the time required for adding culture medium, inoculation, culture induction period, reaction process, discharging, tank washing and empty tank sterilization.
